The stone can be tested with heat. This will not harm it unless the diamond is fake. Real diamonds disperse heat. The test can be carried out at jewellery and diamond merchants
Real diamonds are usually set in quality metals, such as platinum or white gold. The setting and the mount of the diamond should also be checked, as a quality setting is easy to spot.
A real diamond scintillates in different shades of sparkling grey. Other colours or rainbow reflections should be pointed out to the diamond merchant.
A transparency test is also useful when testing a diamond for quality. Turn the diamond upside down and place it over text. If the text can be clearly read through the stone, it is probably not a real diamond.

Do a fog test by breathing on it. The longer the fog remains, the more likely it is to be a fake. A real diamond will disperse the fog very quickly
Place the diamond under a UV light. Real diamonds will reflect a blue fluorescence. This does not apply to all stones, so other types of tests should be carried out.
If the diamond is mounted, you should not be able to see the bottom of a diamond looking directly down from the top
Most importantly, ask to see the diamond's certificate. Every real diamond has been graded by a laboratory, such as GIA for the 4C's, Clarity, Cut, Colour and Carat Weight.
