Sirolimus Belongs To A Group Of Medicines Known As Immunosuppressive Agents - Pillsformedicine

medicinesSirolimus belongs to a group of medicines called immunosuppressants. It helps to control the body's immune system after received an organ transplant. It is used to prevent the body from rejecting the transplanted kidney.

Mechanism of action:

This drug inhibits T lymphocyte activation and proliferation, which occurs in response to antigenic and cytokine stimulation; however, its mechanism is distinct from that of other immunosuppressants. It also inhibits antibody production in cells. This product binds to the immunophilin, FK Binding Protein-12 (FKBP-12), to generate an immunosuppressive complex. The sirolimus: FKBP-12 complex has no effect on calcineurin activity. This complex binds to and inhibits the activation of the mammalian Target of Rapamycin (mTOR), a key regulatory kinase. This inhibition suppresses cytokine-driven T-cell proliferation, inhibiting the phase progression of the cell cycle.

This drug is indicated ...
... for the prophylaxis of organ rejection in adult patients at low to moderate immunological risk receiving a renal transplant. It is recommended that it can be used initially in combination with ciclosporin microemulsion and corticosteroids for 2 to 3 months. This drug may be continued as maintenance therapy with corticosteroids only if ciclosporin microemulsion can be progressively discontinued .

Interactions:

Some medicines may interfere with each other. These include:

cyclosporin used to prevent transplant rejection
antibiotics such as rifampicin, clarithromycin, erythromycin and rifabutin
antifungal medicines such as ketoconazole, clotrimazole, fluconazole and itraconazole
medicines for high blood pressure or heart problems such as diltiazem, nicardipine and verapamil
epilepsy medicines such as carbamazepine, phenobarbitone and phenytoin
medicines for stomach ulcer or reflux such as cimetidine and cisapride
medicines to prevent nausea and vomiting such as metoclopramide
danazol which is used to treat endometriosis
bromocriptine which is used to treat Parkinson's disease
protease inhibitors such as ritonavir and indinavir, which are used to treat HIV/AIDS
St. John's wort.


These medicines may be affected by Sirolimus, or may affect how well it works.

Precautions:

This drug is not recommended for use during pregnancy because it may cause birth defects. Therefore, it is very important that an effective form of birth control be used before starting sirolimus therapy, during this therapy, and for 12 weeks after this therapy has stopped. However, because this medicine may cause serious side effects, breast-feeding is not recommended while you are receiving it.

Kidney Disease, Rash, Fever
Urinary Tract Infection
Fluid Retention in the Legs, Feet, Arms or Hands
High Cholesterol, Stomach Cramp
Low Amount of Potassium in the Blood
Reduction in the Body's Resistance to Infection
Anemia, Joint pain
Decreased Blood Platelets
Incomplete or Infrequent Bowel Movements
Visible Water Retention, Head pain
Feel Like Throwing Up, Diarrhea
High Amount of Triglyceride in the Blood


Storage:

Keep this tablet in their blister pack until it is time to take them.
If you take the tablets out of their blister pack they may not last as well.
Keep this drug in a cool, dry place where the temperature stays below 250C.
Do not store this tablets or any other medicine in the bathroom or near a sink.
Heat and dampness can destroy some medicines. Keep this medicine where children cannot reach it.
