Cold chain testing is how you prove before a shipment leaves that a product will stay inside its temperature range from packing line to patient or plate. Low temperature chambers do the heavy lifting: conditioning components, running the ambient profiles a shipper will face, and simulating the excursions you hope never happen but have to plan for.

What does cold chain testing involve?
Packaging / shipper qualification: loading an insulated shipper with conditioned coolant and a payload, then running summer and winter ambient profiles to confirm the payload holds 2–8 °C (or -20 °C or -70 °C) for the rated duration.
Component conditioning: bringing gel packs, phase-change material and product to a defined start temperature under controlled, documented conditions.
Excursion simulation: deliberately exposing product to out-of-range temperature for a set time to generate data for the excursion assessment procedure.
Freeze-thaw cycling: repeated crossing of the freezing point for biologics and vaccines sensitive to ice formation.
Storage equipment qualification: proving fridges, freezers, ...
... cold rooms and ultra-low freezers hold their range, loaded, before they enter service.
What chamber capability does each task need?
Task Chamber requirement
2–8 °C shipper qualification Programmable ambient profiles (e.g. -10 °C to +40 °C), stable low-temp control, data logging
Frozen (-20 °C) chain Reliable -20 °C to -30 °C operation with even distribution
Ultra-cold (-70 °C) vaccines Ultra-low chamber or -70 °C capable unit; fast pull-down
Freeze-thaw cycling Controlled ramp through 0 °C, repeatable cycle counts
Loaded equipment mapping Not a chamber task — done in situ, but chambers qualify the sensors and loggers used
Why use a chamber instead of a cold room?
Control and profiling. A cold room holds one setpoint; cold chain qualification needs programmed ambient profiles that ramp between extremes on a schedule, tight stability (±0.5 °C) and even distribution so the shipper sees the intended challenge, plus a traceable log for the validation report. A cold test chamber does this down to -40 °C, with a -70 °C option for ultra-cold work; a temperature and humidity chamber adds humidity for packaging and label studies.

Qualifying shippers or simulating excursions? Indeecon's low temperature chambers run programmable ambient profiles to -40 °C (-70 °C option) with validation-ready logging. Request a quote.

How does this connect to your calibration programme?
Every temperature claim in a cold chain study depends on calibrated sensors. The chamber proves the packaging; a NABL-traceable calibration proves the loggers and probes that measured it. Keep interim in-situ checks running with an 8L compact temperature and humidity calibrator so a drifting logger is caught before it invalidates a qualification or triggers a false excursion in the field.

What standards and guidelines apply?
ISTA 7D/7E and ASTM D4169 for thermal transport simulation of insulated shippers.
Good Distribution Practice (GDP) for temperature control, mapping and excursion management.
WHO PQS and national guidance (e.g. CDSCO) for vaccine cold chain.
ICH Q1A storage conditions for the product's own stability basis.
ISO/IEC 17025 traceability for all monitoring sensors and data loggers.
