DNA Fragmentation is the separation or breaking of DNA strands into pieces.

Men with sperm motility defects often have high levels of sperm DNA fragmentation. The degree of DNA fragmentation in sperm cells can predict outcomes for an In Vitro Fertilization and Intracytoplasmic Sperm Injection.

The sperm chromatin dispersion test and TUNEL assay are both effective in detecting sperm DNA damage. Its main units of measurement is the DNA fragmentation Index. A DFI of 20% or more significantly reduces the success rate after ICSI.

Advantages of the sperm DNA fragmentation test:-

Provides information that helps in the clinical diagnosis, management and treatment of male fertility

High Rates of sperm DNA fragmentation and pregnancy potential :-

Embryos derived from sperm with highly fragmented DNA have poor prognosis.
DNA fragmentation could result in initiation of apoptosis (natural cellular death) and mutations resulting in blastocyst arrest, miscarriage and abnormalities in the offspring.
Sperms with high DNA fragmentation fertilizing younger oocytes than older oocytes carry a better prognosis of successful pregnancy, as they are much more efficient at DNA repair of defective sperm.

Elevated testicular temperature

Smoking

Alcohol

Stress
